应用荧光逆行标记与FAGLU荧光组化相结合技术,探索了大白鼠伏隔核5-HT能传人纤维的脑干起源。结果表明,伏隔核5-HT能传入纤维主要来源于同侧中央上核(B_6)、中缝背核(B_7)、中缝正中核(B_8)心其外侧的网状结构(B_9)内的5-HT细胞;此外,在B_6、B_7中,尚存在少量向双侧伏隔发核出分叉投射的5-HT细胞。
Fluorescent retrograde labeling combined with the FAGLU method has performed in 42 rats to demonstrate the origins of the afferent 5-HT fibres of the nucleus accumbens (acb) in the brain stem. In the ipsilateral group, fluorescent tracer PI, DAPI or Bb was injected into acb individually. The animals survived for 2 1/2-3 days (for PI, DAPI) or 12-15 hours (for Bb). Before perfusion via the left cardiac ventricle with the FAGLU solution, colchicine-reserpire-parpyline-L-tryptophan was administered individually to enhance 5-HT fluorescence. The retrograde fluorescent -labeling cells showing specific 5-HT fluorescence were traced under OLYMPUS fluorescence microscope in dried transverse frozen serial sections of the brain stem. The results were as follows: The retrograde fluorescent labeled cells showing 5-HT fluorescence were found in B9, B8. B7, B6. In the bilateral group, PI was injected into acb, and DAPI or Bb was injected into the opposite acb. A few retrograde double fluorescent labeled cells showing 5-HT fluorescence were found in B7 and B6. It was indicated that acb received the afferent fibres of 5-HT cells in B9, B8, B7, B6 in the brain stem. In addition a few of the 5-HT cells in B7, B6 projected their collateral fibres to acb on both sides.
