大鼠移植肾组织急性排斥反应中Fas基因的表达与细胞凋亡的关系

摘要 目的 :探讨肾移植急性排斥反应大鼠移植肾组织中Fas基因的表达与细胞凋亡的关系。方法 :分别取4 6只SD成年大鼠和 4 6只Wistar成年大鼠 ,将SD大鼠左肾原位移植至Wistar大鼠左肾区 ,建立大鼠同种异体肾移植模型 ,以右侧肾为正常对照 ,应用免疫组织化学方法检测移植肾组织Fas基因的表达 ,应用原位DNA片段未端标记法检测移植肾组织细胞凋亡状态。结果 :正常大鼠肾Fas基因主要呈弱阳性或阴性表达 ,而移植肾Fas基因表达显著增强 ,且与肾曲小管细胞凋亡指数呈显著的正相关 (rs=0 .5 1 2 ,P <0 .0 0 1 )。结论 :大鼠移植肾Fas基因的过量表达可引起肾组织细胞的凋亡 。 Aim:To investigate the relationship between Fas gene expression and cell apoptosis during acute rejection in rat kidney allograft. Methods: Rat renal allograft model was contructed. Immunohistochemistry was used to detect the expression of Fas gene. In situ labeling apoptotic DNA fragment detection(TUNEL) was used to detect renal allograft tubule cell apoptosis.Results: Fas protein was negative or weakly expressed in normal rat kidney, whereas Fas protein expression was significantly increased in renal allografts during acute rejection. There was a positive correlation between Fas gene expression and renal allograft tubule cell apoptosis( r s=0.512, P <0.001).Conclusion: Overexpression of Fas gene may lead to renal allograft tubule cell apoptosis, renal structure injury and loss of function.
