摘要
探讨核因子-κB(NF-κB)在小鼠胚泡着床过程中是否存在的短暂激活。方法:采用免疫组织化学检测小鼠子宫内膜NF-κB的亚基p65蛋白的定位和表达,Western印迹法检测子宫内膜NF-κB的抑制性蛋白IκBα的降解。结果:p65在小鼠胚胎着床前后子宫内膜均有表达,其部位主要在腔上皮细胞和腺上皮细胞的胞浆部分,基质细胞和子宫肌层也有微弱表达。妊娠后子宫内膜p65的表达增加,妊娠d5达最高值,d8仍然维持较高水平;而d5IκBα的降解最明显。结论:NF-κB可能通过其短暂激活来参与调节小鼠胚泡着床。
Objective To characterize the expression of the subunit p65 of NF-κB and inhibitor kappa B alpha(IκBα)in mouse uterus during peri-implantation,and therefore to investigate whether transient activation of nuclear factor-κB(NF-κB)occurs during embryo implantation in mice.Methods:Immunohistochemistry technique was used to investigate the expression and localization of p65 in endometrium or decidua,and Western blot analysis was used to detect the levels of IκBαprotein in mouse endometrium or decidua.Results:P65 protein can be detected in stromal cells,epithelial cells of endometrium and in myometrium as well.Staining was predominately seen in the cytoplasm of the cells.Staining intensity for p65 was stronger in the epithelial compartment than in the stromal compartment and myometrium.Staining intensity increased slightly during pregnancy,and it reached a peak in d5.In contrast to p65,the IκBαlevel was the lowest in d5 in the whole groups studied.Conclusion:NF-κB may regulate embryo implantation by its transient activation in mice.
