电针对脊髓背核突触性终末可塑性的影响——电镜定量研究 被引量:7

THE EFFECT OF ELECTRIC ACUPUNCTURE ON THE PLASTICITY OF THE SYNAPTIC TERMINALS IN CLARKE'S NUCLEUS AFTER PARTIAL LUMBOSACRAL DEAFFERENTATION OF THE CAT SPINAL CORD A QUANTITATIVE ULTRASTRUCTURAL STUDY

摘要 实验采用切断猫一侧L_1-L_5、L_7-S_2脊神经背根,保留L_6背根(备用根)模型,研究电针刺激足三里与悬钟、伏兔与三阴交两组穴位30天后对背核可塑性的影响。应用体视学方法测知背核及其神经毯面积均未缩小的情况下,在电镜下比较了两侧背核神经毯单位面积内三种突触性终末的数量变化。发现:针刺的手术侧来自背根有髓纤维的巨大终末数恢复到非手术侧的正常水平,与本室另一组非针刺资料(单纯备用根,其手术侧巨大终末数为正常数量的75%)比较,巨大终末数有显著性增加。表明电针刺激能促进同侧备用根有髓纤维的侧支出芽和突触重建。非背根来源的小扣型终末数在手术侧虽有增多,但其增加幅度与单纯备用根手术侧比较无显著差异。说明针刺对小扣型终末的数量无明显影响。 Five adult male cats with unilateral L1-L5 and L7-S2 dorsal rhizotomy and spared L6 were studied, After operation, the rhizotomy side of animals were subjected to acupuncture at Zusanli, Xuanzhong; Futu and Sanyinjiao points located within the innervating area of L6 spinal nerve of hind limb for two courses. All animals survived for 30 days after operation were perfused with fixative and the L6 segments were dissected. The transverse semithin and ultrathin sections containing Clarke's nucleus of both sides in the same plane were cut. The areas of Clarke's nucleus and neuropil were measured with stereological method under LM, there was no statistically significant atrophy of the areas occupied by Clarke's nucleus and neuropil in rhizotomy side. Quantitative ultrastructural examination was used to observe the plasticity of three types of synaptic terminals in the neuropil of Clarke's nucleus The results showed: (1) The number of giant axon terminals(GATs)originating from dorsal roots in the operated sides (acupunctured sides) returned to normal (control) evel. This suggests that the spared root could sprout and reinnervate to replace the lost synoptic terminals. Compared with the unacupunctured spared root data by Li and Wu, the number of GATs in acupuncture cat increased significantly. It means that acupuncture might promote the myelinated nerve fiber from spared root sprouting and reestablishing synoptic connection. (2) The number of small bouton-typed terminals originating from spinal interneurons localized below L4 showed a statistically significant increase in the operated side. This suggests that heterotypic sprouting occurred in the nondorsal root system. However, the numbers between acupunctured and unacupunctured spared root showed no siginificate difference. It means, acupuncture do not promote the heterotypic sprouting.
