人神经生长抑制因子的单克隆抗体制备及抗原决定簇的研究 被引量:1

摘要 神经生长抑制因子(FIF)是一种特异存在于哺乳动物脑中的金鹰硫蛋白 (MT),又称MT-Ⅲ,具有特异神经细胞生长抑制效应.MT-Ⅲ与MT有70%的序列 同源性,但MT却无此功能.在患AD症病人的大脑中,GIF和mRNA的含量均显著降 低.为了进一步研究,MT-Ⅲ的结构与功能及与AD症的关系,制图示GIF的单克隆抗 体,用ELISA方法研究了GIF,α-结构域和β-结构域,N-末端六肽和N-末端+肽分别 与GIF的单克隆抗体的相互作用,并对单克隆抗体的抗原决定簇进行了分析.结果显示 了GIF的抗原决定簇与其一定的空间构象有密切的关系. Neuronal growth inhibitory factor (GIF), a brain-specific metallothionein protein (Metallothionein- Ⅲ), can inhibit the growth of neuronal cell. Though exhibiting the 70% same sequence, GIF distinguishes from othet MT isoforms by inhibiting neuronal survival in vitro. It was reported that MT- Ⅲ and its mRNA were greatly reduced in Alzheimer's disease (AD) brain. For further research, the monoclonal antibody of GIF was prepared and the interaction between GIF, a-domain. β-domain, peptides and McAb of GIF were studied with ELISA. The epitope of the MT- Ⅲ was studied. The results show that the epitope of GIF is closely related to GIF's conformation.
