新城疫病毒分离株的蚀斑纯化及影响蚀斑形成的主要因素 被引量:23

摘要 从西北地区 1979~ 1999年发生新城疫的鸡群中分离和收集的 15株新城疫病毒 ( NDV) ,经蚀斑纯化 ,共得到 11个克隆毒株 ;对其进行了蚀斑形成能力的测定 ,证明蚀斑形成能力与毒力成正比 ;探讨了温度对蚀斑形成能力的影响 ,证明 NDV蚀斑形成能力在 4 1℃比 37℃强 ,表现为蚀斑出现早、蚀斑大、蚀斑形成单位 ( PFU)高 ,同时在 4 1℃时细胞生长状况良好 ,不易污染 ,故认为在 4 1℃进行 NDV的蚀斑纯化值得推荐。 15 isolates were isolated from chickens of Northwestern China(Shaanxi,Gansu,Xinjiang and Qinghai province) in 1979 1999.11 of them were cloned by plaque test.All 11 isolates could form round and clear plaques on the monolayer cell of embryonat fabricates at 37℃ and 41℃,their sizes and PFU were different.Most of them had a coincided with virulence.The temperature affected the plaque forming.The plaque appeared earlier,bigger and PFU higher at 41℃ than at 37℃;plaque cloning at 41℃ could not affect the virulence NDV strains;There were no variation between 519 bp nucleotides sequence of NDV F gene at 41℃.At the same time,CEF cell grew well and wasn′t easy to be polluted at 41℃.It suggested that plaque purification at 41℃ may be a good method for NDV.
