摘要
将纯化的高表达 1.3copy HBV全基因 ,通过原核显微注射方法 ,建立了近交系 ( Balb/ c)高表达 HBV转基因小鼠模型 ,以血清 HBs Ag为检测指标 ,对该转基因鼠 HBV基因持续表达水平和数代遗传稳定性进行了研究。结果 ,获高表达 HBV( 1.3copy)转基因 Balb/ c小鼠 Founder 2只 ,用回交传代方法 ,本 HBV转基因小鼠已稳定传至第 8代 ,每代转基因鼠阳性率平均保持在 4 4 .6 7% ,性别对 HBV转基因小鼠总体阳性率有影响 ,雄性小鼠的阳性率 ( 4 4.15 % )明显高于雌性小鼠 ( 4 2 .2 6 % ) ( P<0 .0 5 ) ;血清表达 HBs Ag水平较稳定 ,平均为 ( 85 3.2 8± 2 35 .4 3)μg/ L ,雌雄小鼠之间表达水平没有明显差异。
objecet:Inbred 1 3 copy HBV transgenic mice expressing high level HBV were established by microinjection,and the genetic and expressive stability of exogenous gene in the transgenic mice was observed.Methods:Continuous expressing level and passage of HBV transgenic mice were studied by detecting the serum HBsAg in transgenic mice.Results:Two high expressing founders were obtained,and have stably genetics to eighth generation by backcrosses with the average passage rate of 44 67% among offspring per generation.The passage rate in male is higher than in female with the serum HBsAg lever of (453 28±235 42) μg/L.Conclusons:high level expression and stable genetic HBV transgenic mice have been obtained.
