摘要
目的 :检测体外培养的人口腔黏膜上皮异常增生细胞 (DysplasticOralKeritinocyte ,DOK ) ,为口腔癌前病变体外细胞模型建立提供试验依据。方法 :采用细胞计数 ,生长曲线 ,克隆形成率 ,染色体倍性检测等方法 ,比较DOK与正常口腔黏膜上皮角化细胞 (OralKeritinocytes ,OKC)及Tca8113 (舌鳞癌细胞 )细胞体外培养的生物学特性。结果 :DOK在生长速度 ,克隆形成率 ,染色体异倍体率均介于正常细胞与肿瘤细胞之间。结论 :DOK细胞属正常细胞向癌转变的中间过渡型细胞 ,可作为研究口腔癌前病变的细胞模型。
Objective:To study the biological characteristics of Dyspastic Oral Keritinocytes (DOK) in vitro to offer some database for establishing oral pre-malignant cell model in vitro. Method:growth curve, cloning efficiency and DNA aneuploid rate were studied to eveluate the proliferate ability and malignant transform of DOK, oral normal keritonocytes(OKC) and Tca8113 cells.Result:the proliferate ability and malignant transform of DOK are higher than that of OKC but lower than that of Tca8113 cells.Conclusion:DOK is a kind of special transitional cells type from normal to tumor cells, which present a good cell model to study carcinogenesis and chemoprevention.
