摘要
目的 :探讨二陈汤理气化痰抗衰老的作用机理。方法 :采用D 半乳糖小鼠亚急性衰老模型。小鼠颈背部皮下注射D 半乳糖 4 0天 ,中药组注射D 半乳糖同时以 6 32g/ (kg·d-1 )浓缩二陈汤煎液灌胃 4 0天 ,并以空白组作对照 ,观察二陈汤对小鼠脑、胸腺、脾、肝、肾指数、自由基代谢指标血清超氧化物歧化酶 (SOD)、丙二醛 (MDA)及小鼠大脑额前页神经元超微结构的影响。结果 :D 半乳糖诱导的亚急性衰老小鼠脑、胸腺、脾、肝和肾的重量指数明显下降 ,SOD活性明显下降 (P <0 0 1 ) ,MDA含量明显提高 (P <0 0 1 ) ,神经元细胞线粒体肿胀、嵴断裂、空泡样变 ,内质网肿胀、空泡样变 ,核膜模糊或核周间隙扩大 ,核蛋白破坏 ,脱基质 ,核仁边移甚至断裂 ,脂褐素大小形态不规则等衰老改变 ,二陈汤能够提高实验动物脑、胸腺指数 (P <0 0 5 ) ,提高血清SOD(P <0 0 5 )活性 ,降低MDA含量 (P <0 0 1 ) ,使神经元细胞变性程度减轻。结论 :二陈汤具有一定的抗衰老作用 ,并为“痰浊衰老相关性学说”
Objective: To explore the mechanism of two old drugs decoction which can negulat qi to resolve and dissolve phlegm and resist geriatrics. Methods: Use the subacute aging model in mice with D gal.SC the neck and back of mice by D gal(120mg/kg) for 40days,SC the herbs group by D gal let them drink the concentrated two old drugs decoction for 40 dyas,and compared with the empty group,than observe the effect on the indices of the brain,dthymus,spleen,liver and kidney,the activity of SOD of serum,MDA and the ultrastructure of macrophage of the peptidergic neurons. Result: The indices of the subacte aging model in mice with D gal such as brain,thymus,splcen,liver and kidney discrease,the activity of SOD,discreases obviously ( P <0 01),MDA increase obviously( P <0 01),the mitochondria of peptidergic neurons tumid,the cristae cut off,megamitochondria,ER(endoplasmic reticulum) tumid,megamitochondria,nuclear envelope blurred or perinuclear space enlarge,nuclear protein is destroyed,the nuclear matrix drop,nucleolus move away and cut off,the morphological change of the(Lipofucin)LPF,etc.two old drugs decoction can increase the indices of the brain,thymus of the modal mice,increase the actively of the SOD,decrease the MDA and the destroyed of the nucleolus organizer. Conclusions: Two old drugs decoction can resist geriatric change.Meanwhile these result provide reference data the theory of 'Phlegm related to geriatrics'.
