摘要
目的 探讨细胞毒淋巴细胞浸润与鼻咽癌细胞凋亡之间的关系。方法 收集未经治疗的鼻咽癌活检组织30例,采用TUNEL法定量测定鼻咽癌细胞凋亡的程度,用免疫组化的方法检测肿瘤内TIA-1阳性细胞毒淋巴细胞数。结果 按组织学分类,角化性鳞状细胞癌6例,分化型非角化性癌8例,未分化癌16例,平均凋亡指数分别为58.8、62、4和31.6。角化性鳞状细胞癌、分化型非角化性癌与未分化癌之间差异有显著性(P<0.05);每1000个癌细胞范围内浸润的TIA-1阳性细胞为8～62个,在角化性鳞状细胞癌中平均为10.2个,分化型非角化性癌中平均为20.6个,未分化癌平均为22.4个,角化性鳞状细胞癌明显低于分化型非角化性癌、未分化癌,差异有显著性(P<0.05);在分化型非角化性癌、未分化癌中,TIA-1阳性细胞多者,凋亡指数较高(r=0.4047,P<0.05)。结论 鼻咽癌细胞凋亡程度在不同组织学类型中差异有显著性;鼻咽癌组织内TIA-1阳性细胞数在不同的组织学类型中有差别;分化型非角化性癌组织中,TIA-1阳性细胞多者凋亡指数较高。说明细胞毒淋巴细胞具有促进鼻咽癌细胞凋亡的作用。
Objective To investigate the relationship between cytotoxic lymphocytes and apoptosis of nasopha-ryngcal carcinoma cells. Methods Biopsy materials from 30 cases of nasopharyngeal carcinoma(NPC) were collected.The apoptotic index (AI)was evaluated by TUNEL method.TIA-1 positive cytotoxic lymphocytes were detected by immunohistochemisty. Results Thirty cases of NPC were classified as 6 cases of keratinizing squa-mous cell type(KS),8 nonkeratinizing carcinoma (NKC) and 16 undiferentiated carcinoma(UD). Their Als were 58.8,62.4 and 31.6 respectively. The differences of AIs between KS and UD,NKC and UD were signifi-cant( P <0. 05) . The number of tumor - infiltrating TIA - 1 positive cytotoxic lymphocytes was from 8 to 62 in one thousand tumor cell; it was less in KS(mean 10.2) than in NKC-(mean 20. 6) and UD (mean 22.4) significant(P <0.05). In nonkeratinizing NPC,high AIs were related to high number of cytotoxic lymphocytes (r = 0.4047, P <0.05). Conclusions AI is significantly various in different histological subtypes of NPC. NPC with a high number of cytotoxic lymphocytes has a high level of AI. It is suggested that apoptosis in NPC is related cytotoxic lymphocytes.
