The Expression of Transcription Factor GATA-1 and GATA-2 Genes in Bone Marrow Stromal Cells from Patients with Aplastic Anemia or Normal Controls

摘要 目的 探讨转录因子GATA - 1和GATA - 2基因在再障和正常骨髓基质细胞中表达的情况 .方法 体外扩增培养骨髓基质细胞 ,运用RT -PCR -ELISA方法检测GATA - 1和GATA - 2基因的表达情况 ,并对其相对表达水平进行半定量比较 .结果 GATA - 1和GATA - 2基因在正常和再障的骨髓基质细胞中均有一定的表达 .再障的骨髓基质细胞中GATA - 1的表达水平较正常低 ;而GATA - 2的表达水平与正常相比无显著性差异 .正常和再障的骨髓基质细胞中均以GATA - 2的表达占优势 .结论 转录因子GATA - 1和GATA - 2基因可在正常和再障骨髓基质细胞中表达 ,并可能影响骨髓微环境的造血调控作用 。 Objective To investigate the expression of transcription factor GATA-1 and GATA-2 genes in the bone marrow stromal cells(BMSCs)from patients with aplastic anemia (AA) or normal controls. Methods The expression of GATA -1 and GATA-2 genes were analyzed by using RT-PCR-ELISA in the BMSCs from 34 normal cases and 9 cases with AA. The semi-quantitative expression levels of GATA genes in the BMSCs from patients with AA were compared with normal controls. Results The expression of GATA-1 and GATA-2 genes could be detected in the BMSCs from both normal controls and the cases with AA. The expression level of GATA-1 gene in BMSCs from AA was significant lower than the normal controls (p<0.05). It was no difference in the expression level of GATA-2 gene within the BMSCs from normal controls and AA. The dominant expression of GATA-2 gene was found in the BMSCs from AA or normal controls. Conclusions The expression of GATA-1 and GATA-2 genes could be identified in the BMSCs from normal controls and patients with AA. It may influence the regulation of hematopoiesis in the BMSCs and play the role in pathogenesis and development of AA.
