大剂量 HBsAg 疫苗对 HBV 转基因小鼠细胞免疫调节的实验研究 被引量:2

摘要 目的研究大剂量 HBsAg 疫苗对 HBV 转基因小鼠(Tg 鼠)细胞免疫应答影响。方法用大剂量HBsAg 疫苗免疫 Tg 鼠后,用流式细胞术、氚-胸腺嘧啶核苷(^+H-TdR)掺入法和 ELISA,分别检测其树突状细胞(DC)、T 淋巴细胞增殖及其诱生细胞因子 IL-2、IFNγ的水平。结果大剂量 HBsAg 疫苗组 DC 表面共刺激分子和 I-E^k 表达,特异 T 淋巴细胞增殖能力及其诱生细胞因子 IL-2、IFNγ的水平显著高于对照组。结论大剂量HBsAg 疫苗增强 Tg 鼠细胞免疫应答,恢复对乙型肝炎表面抗原的应答。 Objective To study the cellular immune response to the large-dose HBsAg vaccine in transgenic(Tg) mice.Methods After the Tg mice was immunized with HBsAg vaccine,the DC,the level of proliferation and the produc- tion of IL-2 and IFNγ were determined.Results The expression levels of common stimulating factors and I-E^k,the proliferation and the production of IL-2 and IFNγ in experimental group were significantly higher than those in control group.Conclusion HBsAg vaccine can increase the cellular immune response in Tg mice and reconstruct immune re- sponse to HBsAg.
