C_(57)BL/6近交系小鼠Lewis肺癌动物模型:建设性描述肺癌损害程度的指标 被引量:23

摘要 目的:建立肺癌动物模型,探讨肺癌的致病因素,发病机制及防治措施。方法:将传代培养的Lewis肺癌细胞接种于C57BL/6小鼠腋部皮下,制备肺癌模型。观测最佳接种剂量,成瘤率,发瘤平均潜伏期,平均存活时间,平均瘤重,肿瘤体积动态变化,彩色多普勒能量图(CDE)检测瘤内血流丰富程度及病理组织学观察。结果:最佳接种剂量为1×106个细胞,成瘤率80%,发瘤平均潜伏期为(6.3±0.4)d;平均存活时间为(28.0±2.5)d;平均瘤重(9.13±0.03)g;CDE:70%为Ⅲ级,30%为Ⅳ级,频谱(PS)为75.30cm/s,阻力指数(RI)为0.45。结论:Lewis肺癌细胞诱发肺癌模型在实验肺肿瘤研究中可作为首选造模方法。 AIM:To establish the animal model of lung carcinoma so as to investigate its e tiological factor, pathogenesis and measures of prevention and cure. METHODS:The subculturing Lewis lung cancer cells(LLC) were inoculated subcutan eously to the axilla of C57BL/6 mice to make models .The models were observed th e optimum dosage, the rate of forming tumor,the average incubation period of for ming tumor, stock time, tumor weight and development change of tumor volum. The angiogenesis in tumors were detected with color doppler energy(CDE).The changes of pathologic histology were observed. RESULTS:The optimum dosage cells was 1×106, the rate of forming tumor was 80 %, average incubation period,stock time,tumor weight was (6.3±0.4) d,(28.0±2. 5)d,(9.13±0.03) g,respectively.CDE:Grade Ⅲoccupied 70%and Ⅳoccupied 30%.PS was 75.30 cm/s.Resistent Index (RI) was 0.45.
