摘要 白细胞介素18(Interleukin18,IL18)是一种能诱导IFNγ产生的新型细胞因子,在调节Th1型细胞免疫应答中起重要作用。采用逆转录PCR从活化的小鼠腹腔巨噬细胞中获得了小鼠IL18(mIL18)cDNA,测序鉴定正确的片段经EcoRI酶切后克隆入GST融合表达载体pGEX2T,再转化至大肠杆菌DH5α高效表达。 ? Interleukin18(IL18) is a novel potent inducer of IFNgamma production and plays an important role in Th1 type responses.In this study,mouse IL18(mIL18) cDNA was cloned from activated mouse peritoneal macrophages through reverse transcription(RT)PCR.After confirmed by sequencing,mIL18 cDNA was inserted into fusion expression vector pGEX2T at EcoRI site,then transformed into E.coli DH5α.mIL18 fusion protein was purified with high bioactivity efficiently.
