摘要
白细胞介素18(Interleukin18,IL18)是一种能诱导IFNγ产生的新型细胞因子,在调节Th1型细胞免疫应答中起重要作用。采用逆转录PCR从活化的小鼠腹腔巨噬细胞中获得了小鼠IL18(mIL18)cDNA,测序鉴定正确的片段经EcoRI酶切后克隆入GST融合表达载体pGEX2T,再转化至大肠杆菌DH5α高效表达。

Interleukin18(IL18) is a novel potent inducer of IFNgamma production and plays an important role in Th1 type responses.In this study,mouse IL18(mIL18) cDNA was cloned from activated mouse peritoneal macrophages through reverse transcription(RT)PCR.After confirmed by sequencing,mIL18 cDNA was inserted into fusion expression vector pGEX2T at EcoRI site,then transformed into E.coli DH5α.mIL18 fusion protein was purified with high bioactivity efficiently.
