坐骨神经震荡伤后腰髓一氧化氮及凋亡相关蛋白表达的变化 被引量:2

Changes of expression of nitrogen monoxide, Bcl-2 and Bax of the lumbar spinal cord following sciatic nerve injury by high speed firearms

摘要 目的:探讨坐骨神经高速弹丸震荡伤后腰段脊髓一氧化氮(carbonmonoxide,NO)含量和一氧化氮合酶(nitricoxidesynthase,NOS)活性及凋亡相关蛋白Bcl-2、Bax的表达变化及在细胞凋亡发生中的作用。方法:65只大耳白兔分为正常对照、高速弹丸震荡伤组(震荡伤组)和切割伤组。震荡伤组致伤靶点为兔右大腿外侧坐骨神经体表投影线中点,切割伤组在同一致伤水平切断坐骨神经。行腰段脊髓细胞凋亡定性定量检测、Bcl-2和Bax表达半定量分析、脊髓NO含量和NOS活性检测。结果:震荡伤组伤后1,2周可见脱氧核糖核酸末端转移酶介导的缺口末端标记(TUNEL)阳性运动神经元和胶质细胞,伤后3d,1,2周脊髓NO含量和NOS活性升高;伤后1dBcl-2高表达,伤后3dBax高表达。结论:震荡伤组细胞凋亡发生早、数量多,NO和NOS及Bcl-2、Bax表达变化与其发生有一定关系。 AIM:To explore the changes of nitrogen monoxide(NO),nitricoxide synthase(NOS), Bcl-2 and Bax of the lumbar spinal cord after sciatic nerve injury by high spee d firearms in rabbit. METHODS:Sixty-five rabbits were divided into normal group, firearms group and cut group.In firearms group, the target was the midpoint of the lateral body su rface projection of the right sciatic nerve. The sciatic nerve was cut at the sa me level in the cut group. Cells apoptosis was investigated with the technology of DNA gel electrophoresis, flow cytometry and terminal deoxynucleotidyl transfe rase TdT-mediated biotin dUTP nick end-labeling (TUNEL) staining. The content of NO,activity of NOS,expression of Bcl-2 and Bax of the lumbar spinal cord wer e detected also. RESULTS:In firearms group, at the 1st and 2nd week,the apoptosis of the motone urons and glial cells occurred. The content of NO and activity of NOS increased at the 3rd day and the 1st, 2nd week respectively. The expression of Bcl-2 and Bax were high at the 1st and 3rd day respectively.
