摘要
以葡萄幼叶为材料,分别采用溴化十六烷基三甲基铵(CTAB)法、高盐低pH法、简易法、高盐沉淀法和改良十二烷基硫酸钠(SDS)法5种方法提取总DNA,通过A260/A280值、琼脂糖凝胶电泳和RAPD对5种方法所得DNA溶液进行定量、定性分析和综合比较。结果显示高盐低pH法、高盐沉淀法综合效果较好,改良SDS法所得DNA纯度最佳。因此,在实际工作中还应根据实验目的选取最适合的方法。
Five methods,i.e. CTAB,high saltlow pH,basic method,high salt precipitation, improved SDS method, were used to extract total DNA from the young leaves of grape. The resulted DNA samples of five methods were tested qualitatively and quantitatively using the value of A260/A280 、agarose gel electrophoresis and RAPD. The results show that high salt precipitation and high saltlow pH methods are better as a whole. The purity of extracted DNA by improve SDS is the best. So we should select the proper method according to the objetive in the study.
