摘要 目的 :探讨活性氧对K5 6 2细胞凋亡的影响。方法 :用过氧化氢 (hydrogenperoxide ,H2 O2 )作用于对数生长期的K5 6 2细胞。结果 :Hoechst 332 5 8染色法观察到K5 6 2细胞明显凋亡 ;caspase活性定量测定及WesternBlotting检测显示caspase 3, 8, 9同时被激活。结论 :H2 O2 可诱导K5 6 2细胞凋亡 ,且通过同时激活线粒体途径和死亡受体途径起作用。 Objective To investigate the effect of reactive oxygen species on the apoptosis of K562 cells. Methods K562 cells were treated with hydrogen peroxide (H 2O 2 ) of different doses for different time. Results The apoptosis of K562 cell was observed through the Hoechst 33258 staining. The caspase activity determination and Western blotting showed that caspase 3, 8, 9 were all activated. Conclusion Hydrogen peroxide can induce K562 cell apoptosis through both mitochondria and death receptor signal pathways.
