摘要
目的 :观察三氧化二砷 (As2 O3)诱导 K5 6 2细胞凋亡过程中 Caspase3活性的变化。方法 :K 5 6 2细胞分别用浓度为 0 .5μm ol/ L ,1μmol/ L ,2μm ol/ L ,5μmol/ L的 As2 O3处理不同时间 (0 ,6 h,12 h,2 4h,48h,72 h) ,用流式细胞仪检测 K5 6 2细胞凋亡率 ;荧光发光光度计检测 Caspase3活性。结果 :1μm ol/ L~ 5μm ol/ L的 As2 O3可诱导 K5 6 2细胞凋亡 ,As2 O3浓度在 2μm ol/ L以上时作用更明显。 K5 6 2细胞凋亡过程中 Caspase3活性明显升高。结论 :As2 O3可诱导 K5 6 2细胞凋亡 ,以 2μmol/ L以上的浓度作用更明显 ;Caspase3活化参与了 As2 O3诱导 K5 6
Objective:To observation changes of caspase_3 activity in apoptosis of K562 cells induced by arsenic troxide.Methods:K562 cell line were treated with As_2O_3 of 0.5,1,2,5 μmol/L in concentration of 0,6,12,24,48,72 hours respectively.Flow cytometry was used to detect the apoptosis.Spectrofluorometry was used to detect caspase_3 activity.Rusults:K562 cells could be induced to undergo apoptosis by 1 μmol/L~5 μmol/L As_2O_3,especially when the concentration of As_2O_3 was higher than 2 μmol/L.Caspase_3 activity of K562 cells was remarkably increased after the As_2O_3 treatment.Conclusion:The K562 cells can be induced to apoptosis by 1 μmol/L~5 μmol/L As_2O_3,especially when the concentration of As_2O_3 is higher than 2 μmol/L.Caspase_3 activation participates in the processes in inducing the apoptosis of K562 cells.
