扶正化瘀胶囊对肝星状细胞激活的干预 被引量:17

Inhibiting Effect of Fuzhenghuayu Capsule on the Activation of Hepatic Stellate Cells in Rats

摘要 目的 :观察扶正化瘀胶囊 (319方 )对肝星状细胞 (HSC)激活的抑制作用。方法 :1制备 319方含药血清 ,温育原代 HSC,并以正常血清为对照 ,观察 HSC的增殖与 I型胶原分泌。 2用含药血清分别添加于损伤肝枯否细胞与传代 HSC中培养 ,制备含药血清作用过的损伤肝枯否细胞条件培养液与传代 HSC条件培养液 ,并以添加正常血清的条件培养液作为对照 ,温育 HSC,观察 HSC的增殖与 I型胶原分泌。3测定枯否细胞条件培养液中转化生长因子 -β(TGF-β)、血小板衍生生长因子 (PDGF)、表皮生长因子 (VEGF)及传代 HSC条件培养液中 VEGF。结果 :1含药血清能明显抑制原代 HSC的增殖与 I型胶原分泌。 2损伤肝枯否细胞与传代 HSC可明显促进原代 HSC的增殖与 I型胶原分泌 ,含药血清可抑制这一促进作用。 3损伤肝枯否细胞 TGF-β、PDGF、VEGF与传代 HSC中VEGF分泌明显增加 ,含药血清可抑制损伤枯否氏细胞 TGF- β、PDGF及传代 HSC中 VEGF的分泌。结论 :319方不仅可直接抑制 HSC的增殖与 I型胶原的分泌 ,并可抑制 TGF- β1 、PDGF、VEGF等细胞因子 ,抑制 HSC激活的旁分泌与自分泌途径 ,这一作用可能是该药抗肝纤维化的机制之一。 Objective:To observe the inhibiting effect of Fuzhenghuayu capsule (Formula 319) on the activation of hepatic stellate cells (HSC) in rats. Methods:1.Serum containing Formula 319 was prepared and cocultured with primary HSC,and the proliferation and type I collagen secretion of HSC was observed. 2. The serum was added into culture medium of Kupffer cell isolated from injured rat livers and passage activated HSC (Myofibroblaste-like cell, MFBC), and the conditioned culture medium was collected and added into primary HSC culture medium to investigate its effects on the proliferation and type I collagen secretion of HSC. 3. TGF-β, PDGF, VEGF in Kupffer cell culture medium and VEGF in MFBC culture medium were tested.Results: 1. Serum with Formula-319 could significantly inhibit the proliferation and type I collagen secretion of primary HSC. 2.The injured liver Kupffer cell and passage HSC could obviously enhance the proliferation and type I collagen secretion of primary HSC, which could be interrupted by the drug serum. 3. TGF-β, PDGF, VEGF in the culture medium of the injured liver Kupffer cells and VEGF in MFBC culture medium increased significantly, which could be inhibited by the serum with Formula-319. Conclusion: Formula-319 could directly inhibit not only the proliferation and type I collagen secretion of primary HSC, but also the secretion of cytokines of TGF-β, PDGF and VEGF, so as to inhibit the paracrine and autocrine activation of HSC, which might be its chief mechanism to resist hepatic fibrosis.
