高效液相色谱法/双电极电化学检测器测定小鼠脑组织中硝基酪氨酸 被引量:1

摘要  用HPLC结合双电极电化学检测器测定小鼠脑组织蛋白质3 硝基酪氨酸(3 NT)含量。采用4 6mm×150mmWaters0DS2柱,10mmol L醋酸钠 10mmol L柠檬酸 0 2%甲醇为流动相,检测器第一电极电位650mV,第二电极电位750mV。流速1mL min。在此条件下,小鼠脑组织蛋白质水解液3 NT得到良好的分离。方法回收率为93±2%,加标及未加标样品的sr分别为2 7%和6 2%。最低检测量为0 1pmol。在0 01~2 00μmol L浓度范围内线性关系良好,相关系数为0 9992。运用这一方法成功地观察到18~20月龄小鼠脑组织蛋白质3 NT含量比4~6月龄小鼠明显增加(p<0 05)。 3nitrotyrosin(3NT)content in mouse brain tissue was detected by HPLC combined with dielectrod chemical detectro.The column used was 46?mm×150?mm waters ODS2.The motive phase was 10?mmol/L sodium acetate/10?mmol/L citric acid/02% methanol.The first electrical potential was 640 mV and the second was 750 mV.The follow rate was 1?mL/min.Under this condition the 3NT of protein in mouse brain hydrolysate could be isolated properly.The recovery was (93±2)%.The sr of the sample with and without standard were 27 and 62 respectively.The minimum detected amount was 01 pmol.It gave a good linear relationship under concentration between 001-200?μmol/L,r=0999 2.With this method it was observed that the protein 3NT content in mouse brain during 18-20 month age higher than that of the 4-6 month age (p<005).
