摘要 目的 建立一种直接测定尿草酸的方法———离子色谱法 ,评价此方法在实验室和临床研究中的价值。方法 搜集 2 4h尿液用浓盐酸酸化保存 ,取 0 .1ml尿液 ,用 0 .3mmol/L的硼酸溶液稀释 ,通过 0 .2 μm的微孔滤膜过滤而注入离子色谱仪 ,检测出尿草酸浓度。 结果 对尿草酸的最小检测限为 0 .2 5 μmol/L ,批内变异系数CV =3 .8% ,日间CV =4.2 %。尿样回收率 96.5 %~10 5 .2 % ,CV =3 .1% ,此法与比色法显著相关 (r =0 .996,P <0 .0 5 )。结论 此方法简单、准确而快速 。 Objective To establish a direct method for determination of oxalate in urine by ion chromatography and evaluate its value in laboratory and clinic research.Methods Twenty four h urine was collected and acidified with concentrated hydrochloric acid for preservation.0.1 ml aliquot of urine was diluted with 0.3 mmol/L boric acid and filtered through a 0.2 μm pore size Millipore filter prior into ion chromarography,then the concentration of urinary oxalate was determined within 15 min.Results The detection limit for urine oxalate was 0.25 μmol/L.The interassay and between assay coefficients of variation were 3.8% and 4.2% respectively.The recovery for spiked urine samples was 96.5% to 105.2% with a CV of 3.1%.This method had a significant correlation with colorimetry (correlation coefficient was 0.981, P <0.05).Conclusion The method was simple,accurate and rapid,and can be used to determine urinary oxalate in laboratory and clinical practice.
