摘要
大鼠孕19～21天的胚胎小脑,分别放在4℃、培养液1640及-15℃加有10%DMSO抗冻剂的培养液1640中。两组材料保存1、3、7天后。分别移植于成年同种大鼠创伤小脑内。均在移植后12天取受体小脑行常规组织学切片观察。结果是移植的脑组织大部分成活,其组织层次及细胞结构正常、清晰、且有细胞分裂。在-15℃、保存7天的胚脑组织移植后脑细胞均变性死亡。表明4℃是短期保存移植用胚脑组织的最适温度;移植用的胚脑组织低温保存是一可行和适用的方法。
Cerebellar tissues of rat embryo(E19-21)preserved at 4℃,—15℃ of different period were used as transplants inside cerebellum of adult homologus Wistar rats,A small cavity was made by sucking with pipette
before transplantation.Routine microscopic examination of the grafts 9-12 days after operation showed the survival of the tissue,which was confirmed by its normal appearing layers and clear structure of nucleus just as same as
those of the normal embryonic ceredellum.It is conclude that low tempera-ture preservation of brain tissue for transplantation is practicable.
