树(鼠句)(Tupaia belangeri chinensis)消化器官和性腺LDH同工酶的电泳分析

ELECTROPHORETIC ANALYSIS OF LACTATE DEHYDROGENASE ISOENZYMES FROM THE DIGESTIVE ORGANS AND SEX GLANDS OF TREE SHREW ( Tupaia belangeri chinensis )

摘要 采用0.5%琼脂糖凝胶电泳方法对云南树鼩的肝脏、胃、小肠、胰腺、睾丸和卵巢组织的乳酸脱氢酶同工酶进行电泳分析.结果表明,在6种不同组中,乳酸脱氢酶均呈现为5种不同分子形式的同工酶.其LDH_4至LDH_1同工酶随电泳迁移率的增大依次趋于阳极端,LDH_5同工酶则趋于阴极端.按分光光度比色定量法对各同工酶组分进行洗脱定量,分别测得6种组织中5种同工酶组分的相对百分含量.通过酶谱分析,提示在6种不同组织中由于A、B两种基因表达活性的差异,其酶谱特征也各不相同.文中对树鼩6种组织的LDH同工酶的H和M亚基百分率及H/M亚基比率进行了统计分析,提示这与各类组织的生物学功能及代谢类型相关. A comparative analysis of zone electrophoresis by 0.5% agarose gel on lactate dehydrogenase isoenzymes in the digestive organs ( liver, stomach, small intestine and pancreas)and sex glands(testis and ovary)of tree shrew(Tupaia belangeri chinensis ) was made. The LDH isoenzymes were separated and stained according to the method of Zheng, Z.X.(1987). The electrophoresis was performed on 0.5% agarose gel thin layer in a pH 8.6 buffer system containing 0.06 mol/L sodium diethylbarbiturate- 0.2 N hydrochloric acid. The isoenzymic activity was visualized with a staining solution consisting of sodium DL-lactate, nicotinamide adenine dinucleotide(NAD), N-methyl-phenazinium methylsulphate ( PMS ) and nitrotetrazolium blue chloride (NBT).The electropherograms of LDH isoenzymes was scanned at λs = 570nm λscanning speed for 20 mm/min. Five different molecular forms of LDH isoenzymes and alterations of isoenzymic patterns were observed of these six tissues metioned above of the tree shrew. The isoenzymic phe-notypes of five molecular forms are LDH1>LDH2>LDH3>LDH4>LDH5 in stomach, testis and ovary, only the phenotype of live were LDH5>LDH4 >LDH3>LDH2>LDH1. The LDH4 to LDH1 isoenzymes migrated towards the anode and LDH5 isoenzyme toward the cathode in the pH 8.6 buffer. The mean values of the 5 components of LDH isoenzymes in six tissues were measured. The percentage of H subunits was calculated with the formula H% = LDH1% + 3/4 LDH2% + 1/2LDH3% + 1/4LDH4%. The H/M subunit ratio was 0.69, 1.40, 1.10,1.15, 1.60 and 1.50 in above mentioned six tissues, respectively.
