电刺激小脑顶核对持续局灶性脑缺血时钙蛋白酶活性影响的研究 被引量:7

The Effect of Fastigial Nucleus Electrical Stimulation on the Activity of Calpain During Permanent Focal Cerebral Ischemia

摘要 目的 :探讨电刺激小脑顶核 (Fastigialnucleus ,FN)对持续局灶性脑缺血钙蛋白酶活性的影响。方法 :建立大鼠电刺激FN及持续局灶性脑缺血模型 ,应用Western印迹半定量 15 0 0 0 0FBDP含量 (表示钙蛋白酶活性 )及尼氏体染色对神经元数量及形态分级评分。结果 :缺血后 1~ 2 4h ,钙蛋白酶活性不断增高 ,电刺激FN可使其活性增高程度显著减低。在缺血后 12h内 ,电刺激FN使神经元形态及数量评分显著下降。结论 :电刺激FN可抑制局灶性脑缺血时钙蛋白酶活性 ,起神经保护作用。 Aim:To investigate the effect of fastigial nucleus (FN) electrical stimulation on the activity of calpain during permanent focal cerebral ischemia. Methods:FN electrical stimulation and middle cerebral artery occlusion (MCAo) model of rats were established. Semi-quantitative Western blotting was used for detecting the content of 15 kDa FBDP (represented calpain activity) and Nissl's staining, for scoring by neuronal number and size. Results:After 1~24 hours of MCAo without FN electrical stimulation, the activity of calpain increased continuously. While in rats with FN electrical stimulation, the increase of calpain activity reduced significantly. The increase of score by neuronal number and size significantly reduced in 12 h of ischemia.Conclusion:It is suggested that FN electrical stimulation can inhibit the activity of calpain during permanent focal cerebral ischemia and protect the neuron from ischemic injury.
