摘要
目的 :从牛角膜提取精制Ⅰ型和Ⅴ型胶原 ,检测胶原的提取精制过程 ,分析其结构特点。方法 :提取在含有蛋白酶的乙酸溶液中进行 ;精制用不同浓度的NaCl溶液盐析 ;电泳分析胶原的制备过程和结构。结果 :在不同温度下提取的Ⅴ型胶原 ,其组分相对分子质量存在差异 ;部分Ⅴ型胶原分子可能存在二硫键结构。结论 :使用不同浓度的NaCl溶液进行多步盐析 ,可使Ⅴ型胶原与Ⅰ型胶原分开。
Objective:To extract typeⅠ and typeⅤcollagens from bovine cornea, examine the preparing process and analyze their structures. Methods:Collagen extraction was performed in acetic acid solution containing pepsin.Salt precipitation was used to purify the collagens, and the preparing process and structure were analyzed by SDS_PAGE.Results:The component molecular weights of typeⅤ collagen were different when extracted at 4℃ and room temperature. Disulfide bonds potentially exist in some of the typeⅤcollagen molecules.Conclusion:TypeⅠand typeⅤcollagens can be isolated by use of salt precipitation with different concentrations of NaCl solution. [
