α辐射致DNA双链断裂的AFM观测 被引量:4

摘要 利用先进的原子力显微镜(AFM)技术及凝胶电泳技术,对5.48 MeV的α粒子辐照后的水中pGEMTl质粒DNA进行分析,通过AFM观测,得到了清晰的DNA分子及α辐射致其双链断裂的直观图,并采用凝胶电泳对辐照前后DNA的损伤机理进行了研究,结果表明DNA链的断裂数目随着粒子照射剂量的增加而增加,并通过测量DNA片段所占比例关系,可获得DNA的片段分布. Using the methods of AFM and gel electrophoresis, purified plasmid DNA was irradiated by 5.48 MeV a particle were investigated. In atmosphere, the intuitive images of DNA molecules and double-strand breaks of DNA molecules induced by a irradiation were obtained using AFM in the tapping mode. In addition, the plasmid DNA form was analyzed by gel electrophoresis technique. With the intensification of irradiation dose, the number of DNA strand breaks increased. Also, we obtained the fragmental distribution by measuring the fraction of DNA fragments.
