反相高效液相色谱-蒸发光散射检测法测定安宫牛黄胶囊中的胆酸 被引量:7

Determination of Cholic Acid in An-Gong Niuhuang Capsules by Reversed-Phase High Performance Liquid Chromatography with Evaporative Light Scattering Detection

摘要 应用反相高效液相色谱 蒸发光散射检测法对安宫牛黄胶囊中的胆酸含量进行了测定。色谱柱用JASCOC18柱(250mm×4 6mmi d ,5μm),流动相为甲醇 0 1%(体积分数)醋酸水溶液(体积比为9∶1),流速1 0mL/min,柱温为室温;蒸发光散射检测器漂移管温度40℃,载气(N2)流速4 0L/min。在上述条件下测得胆酸的进样量为0 50~5 02μg时线性关系良好(r=0 9993),最低检测限达到0 05μg,平均加样回收率为97 1%。该方法快速简捷,精密度高,重现性和准确度良好,可为安宫牛黄胶囊的质量控制提供更为科学的依据。 A method for the determination of cholic acid in AnGong Niuhuang capsule by reversedphase high performance liquid chromatography (HPLC) with evaporative light scattering detection (ELSD) has been established. Cholic acid was successfully separated on a JASCO C18 column (250 mm×46 mm id, 5 μm) at the ambient temperature, using methanol01% acetic acid solution (9∶1, v/v) as mobile phase. The flow rate was 10 mL/min. The drift tube temperature and nitrogen carrier gas flow rate of the ELSD were set at 40 ℃ and 40 L/min respectively. The linear range was 050-502 μg (r=0999?3). The detection limit was 005 μg and the average recovery was 971%. The results of the experiments showed that this method is accurate, rapid, simple and reproducible, and suitable for the analysis of cholic acid. The method is also proposed to be used for the quality control of AnGong Niuhuang capsules.
