摘要
以Tb^(3+)-环丙沙星作为荧光探针,提出了一种新的荧光光度法测量胆汁酸(BA)。在pH=9.20的酸度条件下,BA能使铽(Tb^(3+))-环丙沙星(CIP)二元络合物中Tb^(3+)位于545nm处的荧光强度增强达15倍,且增强的荧光强度与胆汁酸的含量成正比。测量的线性范围为1.00×10^(-7)-9.00×10^(-7)mol L^(-1)和9.00×10^(-7)-2.00×10^(-6)mol L^(-1),检测限分别为5.63×10^(-9)mol L^(-1)和7.63×10^(-8)mol L^(-1)。该方法成功地应用于血清中胆汁酸的测定。
Bile acid (BA) in serum is determined using the luminescent terbium ciprofloxacin bile acid (Tb^3++ - CIP - BA) complex. The characteristic fluorescence of Tb^3+ at 545nm in Tb^3+- CIP binary complex is observed and the fluorescence intensity increases up to 15 - fold if BA is added to the binary complex. The enhanced fluorescence intensity is in proportion to the concentration of BA. The liner range of the developed method is 1.00×10^4-9.00×10^-1mol^-1 and 9.00×10^-7-2.00×10^-6mol L^-1,with detection limit5.63×10^6mol L^-1 and 7.63×10^-8 mol L^-1 of BA respectively using Tb^3+ - CIP as fluorescent probe. Some organic compounds and albumin in serum samples are removed in order to improve the selectivity of the proposed method. The recovery of this method is over the range of 94.0 to 101.3% and the relative standard deviation is 0.70%. The developed method is successfully ap- plied to the determination of BA in serum samples.
