苯丙氨酸解氨酶脂质体的制备及其抗白血病细胞增殖作用的初探 被引量:6

摘要 目的 探讨苯丙氨酸解氨酶 (PAL)脂质体对小鼠白血病L12 10 细胞和人白血病K562 细胞生长的抑制作用。方法 采用改良的逆相蒸发法制备出PAL脂质体 ,应用MTT还原法分别检测PAL脂质体和游离的PAL对L12 10 和K562 细胞增殖的作用。结果 在酶活性相同条件下 ,PAL脂质体比游离PAL有较强的抑制细胞增殖作用 ,对K562 细胞作用 4 8h ,IC50 分别为 0 .90 5U/ml和 1.14 0U/ml,对L12 10 细胞作用 4 8h ,IC50 分别为 0 .95 8U/ml和 1.5 13U/ml。结论 PAL脂质体能抑制L12 10 和K562 白血病细胞增殖 ,并呈时间和剂量依赖关系 (P <0 .0 1)。 Objective The effect of liposomal phenylalanine ammonia-lyase(PAL) on inhibiting the growth of mouse leukemia L_ 1210 cells and human leukemia K_ 562 cells was studied in vitro. Methods The liposome of PAL was prepared by a modified reverse-phase evaporation method. The inhibitory effects of liposomal PAL and free-PAL on leukemia cells were investigated by MTT colorimetric assay separately. Results MTT assay showed different cytotoxic effect between liposomal PAL and free PAL, whereas an enhanced effect was observed with liposomal PAL. IC_ 50 of liposomal PAL and free PAL on K_ 562 cells was 0.905 U/ml and 1.140 U/ml in 48hs. IC_ 50 of liposomal PAL and free PAL on L_ 1210 cells was 0.958 U/ml and 1.513 U/ml in 48h. Conclusion The liposomal PAL showed certain inhibitory effects on the leukemia cells proliferation. The inhibitory effect was relative to time and concentration( Р < 0.01 ).
