摘要
目的 :探讨黄蜀葵花提取物对 L PS诱导的人中性粒细胞分泌 IL- 8的抑制作用。方法 :分离人血中性粒细胞 ,分别与不同浓度的药物孵育 30分钟后 ,加入终浓度为 10μg/ ml的 L PS,37o C 5 %CO2 培养 2 4小时 ,以双抗夹心 EL ISA法测细胞上清液中 IL - 8的含量 ,并以 NBT法做 PMN活化计数。结果 :1AMM提取物能在一定程度上减少 PMN经 2 4小时培养后上清液中 IL- 8的含量 ,并在一定范围内与药物浓度呈正相关 ,当药物浓度为 10 μg/ ml时即有较为明显的抑制作用 ;2 AMM提取物对经 L PS诱导的 PMN活化的抑制作用 ,在 0 .5、1和 6小时时 ,与对照组相比无显著差异。结论 :黄蜀葵花提取物对 L PS诱导的人中性粒细胞分泌 IL- 8具有抑制作用 ,但本实验尚未观察到其具有抑制 PMN活化的作用。
Objective: To study the inhibitory effects of AMM to the secretion of IL-8 of human PMN induced by LPS. Methods: Human PMN was separated and incubated with AMM at different concentrations for 30 minutes, 10 μg/ml LPS was added, the PMN was incubated for 24 hours in a 37 oC and 5%CO 2 incubator. IL-8 in the culture medium was determined by use of ELISA, PMN activation was counted by NBT method. Results: IL-8 was decreased in culture medium in AMM group, and the effect was concentration-dependent in some range. The obvious inhibitory effects could be observed when AMM was 10 μg/ml. At 0.5,1 and 6 hour, there was no significant inhibitory effects to the PMN activation induced by LPS could be investigated. Conclusion: AMM could inhibit the secretion of IL-8 of human PMN induced by LPS, but the inhibitory effects of AMM to Human PMN activation was not detected.
