摘要
通过对小鼠胚胎着床初期子宫组织块的培养及内细胞团的分离,探讨利用该方法获得胚胎干细胞的可能性.我们用这种方法已建立了一株小鼠胚胎干细胞系,传至第10代,AKP染色细胞呈强阳性,RT-PCR证实Oct 4表达强阳性.结果表明,利用这种方法获得胚胎干细胞是可行的.与传统方法相比,这种新方法极大简化了实验步骤,降低了实验技术难度.
In order to investigate the probability of obtaining embryonic stem(ES) cells by cultivating uterus tissue pieces with implantation embryo , the mouse uterus of 1~3 days implantation embryos were sheared and cultivated. Using this method, a strain of mouse embryonic stem cell line with high expression of Oct4 mRNA and AKP was established. The results show that it is feasible to obtain mouse embryonic stem cells by cultivating uterus tissue pieces with 1~3 days implantation embryos. The experiment protocols of new method are simplified and the difficulty of culture technique is lowered compared with traditional method.
