As_2O_3对组织因子、纤溶酶原激活物抑制剂-1和-2表达的影响

Effects of As_2O_3 on Tissue Factor, Plasminogen Activator Inhibitor-1 and -2 Expression in NB4, HL-60 and THP-1 Cells

摘要 目的 探讨三氧化二砷 (As2 O3)在诱导 NB4、HL - 6 0和 THP- 1白血病细胞凋亡过程中对细胞内组织因子 (TF)、纤溶酶原激活物抑制剂 (PAI) - 1和 - 2表达的影响。方法 使用不同浓度的 As2 O3处理 NB4、HL- 6 0和THP- 1细胞 ,以生长曲线观察 As2 O3对细胞体外增殖的影响 ,并以琼脂糖电泳及流式细胞术观察细胞凋亡情况 ;用酶联免疫吸附测定 (EL ISA)方法检测经 As2 O3处理后细胞内 TF、PAI- 1和 - 2抗原含量的变化。结果  11μm ol/ L和 8μmol/ L 的 As2 O3分别作用 NB4和 HL- 6 0细胞 2 4 h后 ,细胞均出现典型的凋亡 ;而 THP- 1细胞在 8μm ol/ LAs2 O3作用 2 4 h后未出现细胞凋亡。 2 1μmol/ L As2 O3下调 NB4细胞内 TF抗原含量 ,与对照组相比差异显著(P<0 .0 1) ,且 TF抗原水平的降低呈 As2 O3剂量依赖性 ;而 2 μm ol/ L As2 O3诱导 NB4细胞凋亡过程中 ,细胞内PAI- 1抗原含量增加 (P<0 .0 5 )。 38μm ol/ L As2 O3诱导 HL- 6 0细胞凋亡过程中 ,细胞内 TF抗原含量增加 (P<0 .0 1) ,但细胞内 PAI- 2抗原含量降低 (P<0 .0 5 )。 4 THP- 1细胞在 8μm ol/ L As2 O3处理 2 4 h后 ,细胞内 TF和PAI- 2抗原含量均增加 (P <0 .0 5和 P<0 .0 1)。结论  As2 O3对 NB4、HL - 6 0和 THP- 1细胞内的 TF、PAI- Objective To explore the effects of arsenic trioxide(As 2O 3)on tissue factor(TF), plasminogen activator inhibitor 1(PAI 1) and PAI 2 expression in NB4, HL 60 and THP 1 cells. Methods Inhibition of the cell proliferation of NB4,HL 60 and THP 1 cells treated with As 2O 3 in different concentrations was observed by use of growth curves. Apoptosis was assessed by DNA electrophoresis and flow cytometric DNA contents analysis. TF,PAI 1 and PAI 2 antigen levels in cell lysates were measured by enzyme linked immunoassay(ELISA). Results ① After 24 hours exposure to 1 μmol/L As 2O 3 or 8 μmol/L As 2O 3, induced apoptosis of NB4 or HL 60 cells was noted. However induced apoptosis of THP 1 cells was not seen after exposure to 8 μmol/L As 2O 3 for 24 h. ② The TF antigen levels were down regulated during the As 2O 3 induced apoptosis of NB4 cells ( P <0.01), and the down regulation effect of TF antigen levels was in the As 2O 3 concentration dependent fashion. Moreover, 2 μmol/L As 2O 3 increased the PAI 1 antigen levels in NB4 cells ( P <0.05). ③ The TF antigen levels were up regulated and the PAI 2 antigen levels were down regulated during the As 2O 3 induced apoptosis of HL 60 cells ( P <0.01 and P < 0 05, TF and PAI 2 respectively). ④ The TF and PAI 2 antigen levels in THP 1 cells were up regulated after the exposure to 8 μmol/L As 2O 3 for 24 h ( P <0.05 and P <0.01, TF and PAI 2 respectively). Conclusion The effects of As 2O 3 on TF,PAI 1 and PAI 2 antigen levels in NB4,HL 60 and THP 1 cells are distinct.
