维生素C增强As_2O_3诱导宫颈癌HeLa细胞凋亡的研究 被引量:3

摘要 目的:探讨维生素C是否能影响三氧化二砷(As2O3)诱导宫颈癌HeLa细胞凋亡,以及它对细胞内端粒酶活性的影响。方法:用噻唑蓝(MTT)法筛选出维生素C的最佳浓度250μmol.L-1,并用该浓度联合不同浓度的As2O3同时处理HeLa细胞后,用光镜、MTT法和流式细胞术研究细胞生长和凋亡的情况,用TRAP-ELISA法检测细胞内端粒酶活性的变化。结果:单独1μmol.L-1As2O3处理过的细胞生长未受到明显抑制,但联合250μmol.L-1维生素C处理细胞,则细胞生长受到抑制,且部分细胞出现凋亡形态学特征。2、51、0μmol.L-1As2O3联合维生素C用药组的细胞凋亡率也比单独用As2O3组明显增高,且出现凋亡的时间提早。端粒酶活性检测结果提示,联合用药组细胞内端粒酶活性均明显比单独用As2O3组低。结论:小剂量维生素C能增强As2O3诱导宫颈癌HeLa细胞凋亡,并能增强As2O3对细胞内端粒酶活性的抑制作用。 Objective To study whether ascorbic acid can enhance arsenic trioxide to induce apoptosis and to inhibit telomerase activity in cervical cancer HeLa cells. Methods After HeLa cells were treated with various concentrations of As2O3 plus 250 μmol·L^-1 ascorbic acid, effects of As2O3 and ascorbic acid on survival and apoptosis of HeLa cells were determined by light microscope, MTr assay, flow-cytometric analysis. Telomerase activity in HeLa cells was determined by TRAP-ELISA. Results There was no marked effect on the survival of HeLa cells after the cells treated by 1 μmol·L^-1 As2O3 However, after 1 μmol·L^-1 As2O3 plus 250 μmol·L^-1ascorbic acid treated HeLa cells, significant growth inhibition and apoptosis characteristics were found. As to the higher concentrations of AszO3 (2,5,10 μmol·L^-1), the apoptosis rate of HeLa cells treated by As2O3 plus ascorbic acid was higher than that of HeLa cells treated by As2O3 alone and apoptosis characteristics were earlier. Meanwhile, there was more marked telomerase inhibition following to As2O3 plus 250 μmol·L^-1 ascorbic acid treatment than As2O3 alone. Conclusion Ascorbic acid enhances arsenic trioxide to induce apoptosis in HeLa cells, which is related to its inhibiting telomerase activity in HeLa cells.
