摘要 急性白血病治疗中存在的主要问题是白血病细胞对化疗药物产生耐药性。这种现象由多种耐药机制所致 ,包括细胞对化疗药物反应所经历的凋亡失败或药物到达及 (或 )影响细胞内目标的失败。本综述重点讨论后一种机制 ,特别是细胞内药物转运耐药机制。有报道ATP结合盒 (ABC)转运体P 糖蛋白 (P gp)与急性髓性白血病预后相关。另外更引人瞩目的是ABC转运体多药耐药蛋白 (MRP)和穹隆转运体肺耐药蛋白 (LRP) A key issue in the treatment of acute leukemia is the development of resistance to chemotherapeutic drugs. Several mechanisms may account for this phenomenon, i ncluding failure of the cell to undergo apoptosis in response to chemotherapy, o r failure of the drug to reach and/or affect its intracellular target. This revi ew focuses on the latter mechanisms, and on intracellular drug transport resista nce mechanisms in particular. Expression of the ATP-binding cassette (ABC) tran sporter P-glycoprotein (P-gp) has generally been reported to correlate with pr og nosis in acute myeloid leukemia (AML). Additionally, of more controversial, expr ession of the ABC transporter multidrug resistance protein (MRP) and the vault- transporter lung resistance protein (LRP) have been correlated with the outcome in AML.
