摘要
目的 研究重组结核分支杆菌 3 80 0 0蛋白诱发豚鼠迟发型超敏 (DTH)反应的特异性及其作为皮试诊断品的可行性。方法 皮肤试剂轮圈法 :将豚鼠分为 5组、每组 5只 ,分别为H3 7Rv全菌体、卡介苗、堪萨斯、偶发、瘰疬分支杆菌致敏备用。生理盐水为阴性对照 ,5IU国际标准品PPD为阳性对照。将 0 2 μg3 80 0 0蛋白质分别注入每只致敏豚鼠的背部双侧皮肤。于注射后 2 4,48h分别测量红肿或硬结的纵、横径 ,并记录两者平均值。结果 PPD与所有受试分支杆菌致敏的豚鼠均产生较强的DTH反应。重组 3 80 0 0蛋白对结核分支杆菌致敏豚鼠所产生的DTH反应与PPD相似 ,而对堪萨斯和偶发分支杆菌致敏的豚鼠不产生DTH反应。结论 3 80 0 0蛋白诱发豚鼠的DTH反应与PPD相似 ,而特异性高于PPD 。
Objective To evaluate the specificity of recombinant 38000 protein (r- 38000 ) of mycobacterium tuberculosis as an antigen to induce delay-typed hypersensitisation (DTH) in guinea pig and its feasibility as a diagnostic reagent. Methods Strains of mycobacterium tuberculosis (H37Rv), BCG, kansasii, fortuitum and scrofulaceum were inoculated into abdomen of guinea pig respectively till being sensitized. Guinea pigs were injected intracutaneously with 0 2 μg of r- 38000 protein for study, 5 IU of PPD as positive control or normal saline as negative control. Skin response was measured by transveres and longitudinal mean diameters 24 and 48 hours after injection.Results Guinea pogs injected with mycobacterium tuberculosis had a positive skin response to r- 38000 protein similar to PPD. Guinea pigs with mycobacterium kansasii, fortuitum, scrofulaceum or BCG had no or very weak response to r- 38000 protein. All guinea pigs had positive response to PPD.Conclusion r- 38000 protein has similar function as PPD to induce DTH in guinea pig, but its specificity to mycobacterium tuberculosis is more obvious and may be used as a new reagent for diagnosis of tuberculosis.
