摘要
目的研究钙离子浓度对大鼠精子自发性顶体反应的影响。方法不同钙离子 (Ca2 + )浓度的EKRB改良培养液含 0 .3%牛血清白蛋白于 37℃ 5 %CO2 的培养箱中培养精子 ,用金霉素染色法检测不同时间精子获能和自发性顶体反应的百分率。观察孕酮和钙离子载体A2 3187诱导精子顶体反应的情况。结果在不同Ca2 + 浓度的EKRB培养液中 ,Ca2 + 浓度为 0 .7mmol/L时 ,大鼠B型精子和自发性顶体反应的百分率均较低 ;1.0mmol/L时 ,其B型精子百分率较高 ,自发性顶体反应的百分率较低 ;1.3mmol/L时 ,其B型精子和自发性顶体反应的百分率均较高。结论EKRB改良培养液中钙离子浓度为 1.0mmol/L时 ,既能使精子较好地获能 ,又能使自发性顶体反应的百分率较低。
ObjectivesTo study the effects of calcium ion concentration on spontaneous acrosome reaction of rat spermatozoa.MethodsThe rat spermatozoa were cultivated in the modified EKRB medium containing 0.3% bovine serum albumin (BSA) and various concentrations of Ca 2+ within 5% CO 2 incubator at 37°C. Chlortetracycline fluorescence assay was employed to detect the percentage of spermatozoa capacitation and spontaneous acrosome reaction . Progesterone induced and Ca 2+ carrier A23187 induced spontaneous acrosome reactions were also observed.ResultsThe percentages of spontaneous acrosome reaction and pattern B spermatozoa were low at 30 min, 60 min and 90 min of incubation when Ca 2+ concentration reached 0.7 mmol/L.The percentage of spontaneous acrosome reaction was rather lower and that of pattern B spermatozoa was higher at Ca 2+ concentration of .1.0 mmol/L. The percentages of spontaneous acrosome reaction and pattern B spermatozoa became all high when Ca 2+ .was elevated to 1.3 mmol/L.ConclusionThe modified EKRB medium containing 1.0 mmol/L calcium can bring about a better capacitation and lower percentage of spontaneous acrosome reaction for rat spermatozoa.
