套式聚合酶链反应在豚鼠巨细胞病毒宫内感染模型中的应用价值 被引量:3

摘要 目的 运用套式聚合酶链反应 (N -PCR)检测豚鼠巨细胞病毒 (GPCMV)宫内感染 ,并探讨N -PCR在模型建立中的应用价值。方法 选择无感染史豚鼠 ,受孕后腹腔接种GPCMV ,分娩后 2 4h内处死 ;观察妊娠结局 ,运用N -PCR检测亲代及子代感染情况 ,并对标本进行病理学检查。N -PCR的特异性实验设阴性、空白和阳性对照。结果 感染孕鼠出现病毒血症征象 ,胎仔出现流产、死胎等异常结局。N -PCR检测显示 ,亲代 10 0 % ( 10 / 10 )感染 ,子代 96 .0 % ( 2 4 / 2 5 )感染 ,且感染脏器均出现非特异性组织病理学变化。N -PCR有较高的GPCMV检测特异性和灵敏性。结论 N -PCR检测GPCMV宫内感染 ,简便、快速、灵敏、准确 。 Objective: To detect the congenital infection with cytomegalovirus in the Guinea pigs using the Nest-Polymerase Chain Reaction(N-PCR), discuss the value of N-PCR using in the model. Methods: Select Guinea pigs without infestation, intraperiloneal inoculation with GPCMV before pregnancy, then kill them after delivery between 24 hours, observe the outcome of the cyesis, detect the infestations of the parental generation and filial generation using N-PCR, and procee a patholog detect with the samples. Negative, vacuity, positive control are set up in the specific detect of N-PCR. Results: Viremia occurred on the maternal animal, abortions, fetal deaths and some other abnormal outcome can be seen in their youngth. The parental generation's infection rate is 100%(10/10) and the filial generation's is 96.0(24/25). All the visceras of the infection's Guniea pig occurrenc non-specific histological change. N-PCR has the specificity and susceptiveness in the detection about GPCMV. Conclusion: N-PCR is a satisfactory method using in the GPCMV congenital Infection's model, it has many virtues like convenient, swif, sensitive and accurate.
