摘要
应用深低温生物技术对正常人(N=4)和白血病患者(N=9)天然杀伤(NK)细胞冻存后,采用^(125)I 释放法,研究 NK 细胞对杀伤肿瘤细胞活性的影响。结果表明使用防冻剂二甲亚砜(DMSO),液氮(-196℃)储存,能保存 NK 细胞活性(冻前:正常人54.7±8.0%,白血病8.4±5.2%,冻后:正常人55.2±7.3%,白血病8.6±4.6%。P 值均分别>0.5。
Natural killer(NK)cells from patients with leukemia were isolated and cryoprese- rved successfully at-196℃ for the study on cytotoxicity of NK cells.Dimethyl-sulfoxide (DMOS)was used as the cryoprotectant.The NK cells were cooled and frozen using the routine method.The cytotoxicity of NK cells was determined by ^(125)I release assay. The NK cells activity of patients is maintained by cryopreservation(before cryopreser- vation:8.4±5.2%;after cryopreservation:8.6±4.6%;P>0.5).
