摘要
目的 :建立糖尿病 ( DM)并发去卵巢大鼠骨质疏松症模型 ,为筛选绝经后老年糖尿病患者骨质疏松症的治疗药物提供动物模型。方法 :雌性 Wistar大鼠 40只随机分成正常对照组 ,DM模型组 [以链脲佐菌素 ( STZ)制备 DM骨质疏松动物模型 ],单纯去势组 (去除双侧卵巢 ,建立绝经后骨质疏松症模型 ) ,DM并发去卵巢大鼠组 (以 STZ制成 DM动物模型后 ,进行“去势”手术 ,制成DM并发绝经后的骨质疏松症动物模型 ) ;测定各组大鼠股骨骨密度、血清雌二醇含量 ;将各组鼠的股骨制成切片进行观察。结果 :DM模型组、DM并发去势组血糖、尿糖显著高于正常对照组及单纯去势组 ( P<0 .0 5 ) ;DM并发去势组与正常对照组、单纯 DM组相比 ,骨密度、雌二醇显著下降( P<0 .0 5 ) ,与单纯去势组相比有所下降 ( P>0 .0 5 ) ;DM并发去势组与各组相比骨皮质明显变薄、骨小梁数量减少、骨组织结构受到破坏最为明显。结论
Objective: To establish the osteoporosis model of castrated rat with diabetes mellitus (DM) and use the model to screen drugs for treating the patients with postmenopausal osteoporosis and DM. Methods: 40 female rats were randomly divided into 4 groups: the normal control group, the DM model group, the castrated rat group and the castrated rat with DM group . The density of femur and content of estrogen were determined and femur slices were made and observed . Results: The content of blood sugar and urine sugar in DM group and castrated rat with DM group were higher than that in control group and castrated group. The density of femur and the content of serum estrogen in castrated rat with DM group reduced, comparing with control group and DM group. Conclusion: The osteoporosis model of castrated rat with DM was successfully established in the study.
