摘要
目的 :探讨血小板CD6 2P在 2型糖尿病大鼠肾脏病变中的变化及意义。方法 :wistar大鼠 2 4只给予高脂高糖饲料喂养 4周形成胰岛素抵抗后腹腔注射小剂量链脲佐菌素 (STZ ,30mg/kg)形成 2型糖尿病模型 ,另12只喂以普通饲料作为正常对照组 ,分别在胰岛素抵抗期、糖尿病成模 12周测二组大鼠血小板CD6 2P的平均荧光强度 (用流式细胞仪 )、尿白蛋白排泄率 (RIA法 )并观察肾脏形态学改变 (光镜 )。结果 :胰岛素抵抗期血小板CD6 2P已有增高趋势 ,但是与正常对照组相比统计学上无显著差异 (2 .89± 0 .10vs.2 .76± 0 .17incontrols ,P >0 .0 5 ) ,此时肾脏形态学、尿白蛋白排泄率无明显变化 ;糖尿病成模 12周时 ,血小板CD6 2P的表达对照组显著增高 ,(5 .2 0± 0 .2 7indiabetes ,vs.2 .6 8± 0 .15incontrols,P <0 .0 0 1)尿白蛋白排泄率无明显变化 ,光镜下肾小球系膜基质明显增多 ,基底膜增厚。结论 :2型糖尿病大鼠肾脏病变过程中 ,血小板CD6 2P的升高早于尿白蛋白排泄率的增高 ,在血小板CD6 2P升高时肾脏已发生了形态学的改变 ,血小板CD6 2P在糖尿病肾病的发生、发展中起重要的作用 ,可作为诊断早期糖尿病肾病更敏感的指标。
Objective:To study the change of platelets CD62P in the different stages of diabetes mellitus rats and its significance in the process of diabetic nephropathy.Methods: 24 wistar rats were fed in a diet enriched in glucose and fat for 4 week to induce insulin resistance(IR) ,then injected intraperitoneally with low dose streptozotocin(30mg/kg)to induce type Ⅱ diabetes ,the other 12 wistar rats were fed on a normal diet as controls .In the insulin resistance stage ,the 12th week after developed diabetes platelets CD62P were determined by flow cytometory(FCM),urinary albumin excretion(UAE)were measured by RIA,and the renal morphologic changes were observed under light microscope .Results:In the IR stage,platelets CD62P had a trend to increase but not significantly(2.89±0.10 vs.2.76±0.10 in controls,P>0.05),meanwile ,urinary albumin excretion(UAE)and renal morphology were not markedly different compared with controls.In the 12th weed after developed diabetes the expressions of platelets CD62P were signficantly enhanced compared with the other group(5.20±0.27 in diabwetes ,vs.2.68±0.15 in controls,P<0.001),and UAE were not significantly different ,but the glomerular mesangium matrix had obviously increased.Conclusion: Prior to UAE increase ,platelets CD62P has significantly increased,meanwile,the renalmorphology has obviously changed .So the expression of pltelets CD62P may be a more sensitive marker in the diagnosis of early diabetic nephropathy ,it may has a role in the development of diabetic nephropathy.
