摘要
结扎大鼠左冠状动脉形成急性心肌梗塞后30min,血小板与白细胞的粘附率较正常大鼠及假手术大鼠显著增加,其值为31.9±5.4%;到16~24h达高峰,粘附率为60%以上。心肌梗塞大鼠的血小板与正常大鼠的白细胞以及正常大鼠的血小板与心肌梗塞大鼠的白细胞的粘附率也明显增加。提示心肌梗塞大鼠的血小板与白细胞粘附率增加,不仅是因为血小板的激活,同时还由于白细胞的激活。血小板与白细胞的粘附需要Ca2+参予,但不需要能量。此外,肝素及硫酸右旋糖苷可抑制心肌梗塞的血小板与白细胞的粘附,而右旋糖苷则无抑制作用。
Interaction between platelets and neutrophils in acute myocardial ischaemia (AMI) rats were studied by observing the formation of rosette from both kinds of cells. The adhesion rate of the sham operated animals were 13.4±3.8%(n=6). 30 min after ligation of left coronary artery,the adhesion rate increased to 31.9±5.4%(n=6,P<0.001).The binding rate increased with time and reached peak after 16~24h of AMI,which was higher than 60%.48h after AMI,the adhesion became to decrease. The adhesion rate of platelets from AMI rats with normal neutrophils or normal platelets with neutrophils from AMI rats were 28.8±6.6% and 29.4±5.9% respectively. This indicated that both platelets and neutrophils were activated and contributed to the increased cells′ adhesion in AMI rats. The results also showed that the interaction between the cells was Ca 2+ dependent. Platelets could still bind to neutrophils even when platelets were fixed or reacted at 4℃. It suggested that no energy was needed in the adhesive process. Heparin (40U/ml) or dextran sulfate (400μg/ml) inhibited cells binding but dextran did not. The adhexive molecule—GMP 140 might be involved in the mechanism of the interaction between platelets and neutrophils.
