摘要
本文利用流式细胞仪研究了雷公藤多甙(TW)对淋巴细胞膜表面标志及对小鼠脾细胞产生白细胞介素2(IL-2)水平和产生抗休效价的影响,结果证实TW在体外可以抑制小鼠的T淋巴细胞,但不影响B淋巴细胞;Tw还能抑制小鼠脾细胞产生IL-2的能力。在体内,TW明显抑制小鼠的T和B淋巴细胞及抗体效价,并且也同样抑制小鼠脾细胞产生IL-2的水平。
To clarify the mechanism of therapeutic effect of TW on glomerular disease, both in vitro & in vivo investigations were made in mice to observe the sup poressive effects fp this drug on various subpopulations of lymphocytes. Spleen cells from C57BL/6 J mice were used for culture study with TW ( 1 μg/ml) added to the media at different wells. It was found that under the stimulating effect of ConA
( 5 μg/ml) , proliferation of Ly_2 (suppressive T cell) and L_3T_4 (helper T cell) were inhibited by this drug, but 6B_2 population (B cell) was apparently not influenced. In vivo study was also conducted in C57BL/6 J mice. TW in the dosage of 30mg/kg was administered per OS with mice spleen cells obtained on 14th day. Sub-populations of lymphocytes including Thy_(1.2). Ly_2, L_3T_4 and 6B_2 were identified and counted with flow cytometry(FACS440). It was found that the values of all these subpopulations of lymphocytes were significantly lower than the controls.
