摘要 本文报道应用含血清培养液及无血清培养液进行细胞融合,共获得8株稳定分泌抗纤维连接蛋白单克隆抗体杂交瘤细胞株。Western Blot证实McAb仅识别220KD的Fn。角膜冰冻切片,McAb经PAP法显色,Fn分布带与文献报道相同。ELISA双抗体夹心法测定18例献血员血浆Fn含量,结果为0.342±0.026mg/ml,与文献报道极为相近,且重复性好。 In this paper, it was reported that 8 hybridoas secreting monoclonal antibodies against human fibronectin had been est blished by serum containing culture or serum-free culture. McAbs reacted with Fn of 220KD by Western Blot. With immunohistochemical PAP enzmye staining in the tissue of cornea the distribution of Fn recognized by McAbs was similar to that of published materials. The content of human blood Fn detected from 18 person was 0.342±0.026 mg/ml by double antibody sandwich-ELISA, which was quite the same with published materials with stable repetition.
