摘要
以肾综合征出血热病毒(HFRSV)人McAb的检测为模型,建立了测定McAb亲和常数的简易方法。其基本原理是在两种不同浓度抗原包被的微板上进行系列稀释的抗体测定,然后比较由此获得的两条S形反应曲线的OD50(50%最大OD值),代入方程式便可得出亲和常数K值。该法简便、迅速、可靠,是筛选、测定B细胞杂交瘤产物的理想方法。
The method for measurement of monoclonal antibody (McAb) affinity constant is reported basing on enzyme linked immuno-sorbent assay(ELISA). Its principle is deter-mining a series of diluted antibody on the microplate coated by different concentration of antigens, then comparing different OD50 (50 per cent greatest optical density)derived different S type of response curves, and calculating affinity constant K by a special equation. The result is suggested that the method, simple,rapid and reliable, can be used in screening and characterization of McAb.
