摘要
作者观察了链脲佐菌素(STZ)糖尿病大鼠肝细胞膜胰岛素受体的改变及胰岛素治疗对受体的影响。结果表明:STZ糖尿病大鼠肝细胞胰岛素受体数目及亲和常数均增高,胰岛素治疗可使其逆转。血胰岛素水平与受体数目呈负相关。胰岛素受体的变化与糖尿病病程无关。
The purpose of this study is to investigate the changes in the number and affinity of insulin receptors on liver plasma membranes of streptozotocin (65 mg/kg)-induced rats before and after insulin treatment. 32 Wister rats (150 - 250 g) were divided into four groups randomly: 72-h untreated diabetic control group, 2-wk untreated diabetic group and insulin-treated diabetic group (NPH insulin 6-8 U/d administered subcutaneously for 11 days).
The results showed that specific insulin binding to liver plasma membranes from diabetic rats was two times higher than those of the control rats. Scatchard analysis revealed that both the receptor number and affinity constant were increased, and could be returned to normal by insulin treatment. There was no difference between the two untreated diabetic groups. Linear correlation analysis of these data showed that serum insulin level was in negative correlation with the number of insulin receptors on liver plasma membranes.
