摘要
本实验将人淋巴毒素(LT)基因5,侧翼704bp顺序插在氯霉素乙酰基转移酶(CAT)基因前构建出pLT704CAT。用此质粒转染四种哺乳动物细胞,通过测定CAT活性对人LT基因上游调控顺序的活性及其组织特异性选择进行了研究。结果表明,人LT基因上游顺序中含有该基因的启动子,通常受负控制因素的影响而不表现活性,有丝分裂原对人LT基因自动子活性的诱导效应有明显的细胞特异性。
In this study, a pLT704 CAT plasmid was constructed by inserting a 704bp fragment of the 5' flanking sequences of human lymphotoxin gene in front of the chloramphenicol acetyl transferase (CAT) gene. There were four mammalian cell lines under transfection with this plasmid, the transcription promoting activity and the tissue specific selection of the 704bp fragment were studied. The results suggest that the promoter is located in the upstream sequences of human LT gene. Its trascriptional promoting.activity is under negative control and the mitogenic induction effect of its activity appears tissue specific.
