丙氨酰-谷氨酰胺对人结肠癌细胞株增殖活性的影响 被引量:3

Proliferative effect of Ala-Gln on human colonic carcinoma cell

摘要 目的 研究丙氨酰-谷氨酰胺(Ala-Gln)对人结肠癌细胞株增殖活性的影响。方法 经体外培养结肠癌细胞,加入不同浓度的Ala-Gln后收集细胞,应用流式细胞技术,检测癌细胞的细胞周期及凋亡情况。结果 随着细胞培养液中的浓度从1.0 mmol/L递增至8.0 mmol/L时,细胞凋亡率从8.94%降至3.78%,Ala-Gln浓度与细胞凋亡率呈负相关(r=-0.880,P<0.01)。加入氟尿嘧啶(5-Fu)后细胞凋亡率从18.01%增至36.01%,Ala-Gln浓度与细胞凋亡率呈直线正相关(r=0.942,P<0.01)。进一步增加Ala-Gln浓度,从10.0 mmol/L递增至45.0 mmol/L时,细胞凋亡率分别为16.89%、15.46%、18.47%、22.08%、18.75%、16.56%、20.89%、15.57%,细胞凋亡率与Ala-Gln浓度变化无直线相关关系。结论 在一定浓度范围内,Ala-Gln可减少结肠癌细胞的凋亡,促进G_0/G_1期的细胞增多,增强化疗药物的作用。 Objective To study the proliferative effect of Ala-Gln on human colonic carcinoma cell line LSI74T. Methods Ala-Gln was added in culture liquids in vitro. The apoptosis and cell cycle were measured by flow cytometry. Results With the concentration of Ala-Gln in culture liquids increased from 1.0 mmol/L to 8.0 mmol/L, the apoptosis rate of carcinoma cell decreased from 8.94% to 3.78% and a negative linear correlation was found between the concentration and the apoptosis rate (r=-0.880, P<0.01). When 5-Fu was added in such culture liquids, the apoptosis rate of carcinoma cell increased from 18.01% to 36.01%. With the concentration of Ala-Gln increased further from 10.0 mmol/L to 45.0 mmol/L, the apoptosis rate had no linear correlation with the concentration(P<0.05). Conclusions Ala-Gln in certain concentration can decrease the apoptosis of carcinoma cell and increase the proportion of cells in C_(0/1) phase, which may enhance the effect of chemotherapy.
