摘要 目的 探讨精氨酸升压素 (AVP)对心肌成纤维细胞 (CFs)核转录因子 (NF κB)活性的影响。方法 胰酶消化法分离、培养新生SD大鼠CFs,分别采用免疫荧光 -共聚焦显微镜法和蛋白质印迹技术检测基础状态下和AVP干预下CFs中NF κB的细胞内定位和CFs核中NF κB的蛋白含量。结果 基础状态下CFs的NF κB主要分布于细胞浆 ,细胞核中无NF κB蛋白表达 ,AVP干预后CFs的NF κB主要分布于细胞核 ,细胞浆中NF κB显著减少。结论 AVP能够激活CFs的NF κB ,使其从细胞浆转位至细胞核 ,提示AVP对CFs的调节作用可能是通过NF κB激活通路发挥的。 Objective To investigate the effects of arginine vasopressin(AVP) on nuclear factor κB (NF κB) activity in cultured cardiac fibroblasts(CFs) Methods CFs were isolated by trypsin digestion method Immunofluorescence interactive laser cytometer techniques was used to estimate the localization of NF κB in CFs Western blotting were used to study the protein expression of NF κB in nucleus of CFs with or without AVP Results NF κB is localized in cytoplasm of CFs in control group and the contents of NF κB in cytoplasm significantly decreased in AVP group NF κB protein was found in CFs nucleus in AVP group which is in sharp contrast to the negative finding in control Conclusion AVP activate NF κB and it mobilize from cytoplasm to nucleus in cultured CFs, suggesting that some regulating effects of AVP on CFs might be mediating NF κB activation pathway
