摘要
目的 观察高蔗糖喂养引起的胰岛素抵抗对肾素依赖性高血压大鼠左室肥厚的影响。方法 Wistar雄性大鼠30只 ,按Kuwajima法制成两肾一夹肾性高血压模型后 ,随机分为A、B两组 ,A组为对照组 ,普通饲料喂养 ,B组为高糖组 ,高蔗糖饲料喂养。三周、八周观察其空腹血糖、空腹胰岛素的水平 ,八周时测其血浆和心肌组织的肾素、血管紧张素Ⅱ、醛固酮水平 ,以及左室相对质量 (LVW/BW)。结果 不同饲料喂养后三周 ,高糖组胰岛素水平明显升高 ,胰岛素敏感指数下降 ,与对照组相比有显著性 (P <0 0 1 )。八周时血浆肾素、血管紧张素Ⅱ水平与对照组相比无显著性(P >0 0 5) ,血浆醛固酮水平较对照组增加 2 8% ,有显著性差别 (P <0 0 5) ;心肌组织中肾素、血管紧张素Ⅱ、醛固酮水平与对照组相比分别增加 2 0 %、30 %、2 3 % ,具有显著性差别 (P <0 0 1 )。高糖组LVW /BW与对照组相比增加2 5 % ,有显著性差别 (P <0 0 5)。结论 胰岛素抵抗促进肾素依赖性高血压大鼠左室肥厚的形成 ,可能与高胰岛素血症引起心肌组织肾素血管紧张素系统 (RAS)
Objective To observe the effects of high sucrose diet on left ventricular hypertrophy in renin dependent hypertensive rats Methods According to Kuwajima method, 30 male Wistar rats were produced renal hypertension model by two kidney one clip All rats were divided into A and B groups randomly Group A was fed with standard chew, group B was fed with high sucrose After 8 weeks, levels of fasting plasma glucose, fasting plasma insulin and renin angiotensin Ⅱ aldosterone in plasma and myocardium were tested Results At 3 rd week plasma insulin level was increased and insulin sensitive index was decreased in group B more significantly than that in group A( P <0 01) At 8rd weeks, while little difference of plasma renin and angiotensin Ⅱ were found between group A and group B( P >0 05), plasma aldosterone increased 28% in group B( P <0 05) Renin, angiotensin Ⅱ and aldosterone in myocardium tissue are increased 20%?30% and 23% respectively in the high sucrose fed rats( P <0 01) with LVW/BW increased 25%( P <0 05) as well Conclusion Insulin resistance facilitated left ventricular hypertrophy in renin dependent hypertensive rats Hyperinsulinism may initiate activation of RAS in myocardium tissue
