高蔗糖膳食诱发大鼠胰岛素抵抗的研究 被引量:10

摘要 用高蔗糖饲料喂养雄性SD大鼠,诱发高胰岛素血症及胰岛素抵抗。实验发现第2周时高糖组血糖明显低于对照组,4周后与对照组相比无明显区别。高糖组血清胰岛素水平从第2周开始升高,4周后升高更为明显(P<001)。饲养20周时取肝,制备肝细胞膜可溶性胰岛素受体,经胰岛素受体放射配体法测定,发现高糖组与对照组间受体亲和力无明显差别,但高糖组的受体最大特异结合与受体浓度低于对照组(P<0.05),说明高蔗糖膳食可引起雄性SD大鼠高胰岛素血症。 We fed male SD rats with high sucrose diet to induce hyperinsulinemia and insulin resistance. The serum glucose concentration of high sucrose diet (HSD) group was lower than that of the control in the 2nd week, serum insulin level of HSD group incresed. Four weeks later, insulin level was getting hihger and higher than that of the control one (P<0.01). We observed that HSD could induce hyperinsulinemia and IR, but no significant change at serum glucose level. The biggest special insulin binding and the concentration of solubilized insulin receptor from rats liver in HSD group were lower than that of the control group (P<0.05), and correlated to serum insulin level, we demonstrated that high sucrose diet can induce hyperinsulinemia and the changes of the kinetics of insulin receptor binding can aggravate insulin resistance in male SD rats.
